黄芪多糖对大黄鱼头肾巨噬细胞的免疫调节作用

摘要:
为探究黄芪多糖(Astragalus polysaccharide,APS)对大黄鱼(Larimichthys crocea)头肾巨噬细胞的免疫调节作用,本研究从植物黄芪中分离纯化得到APS,用流式细胞仪检测其对头肾巨噬细胞呼吸爆发和吞噬活性的影响,并采用qPCR检测APS对3种细胞因子TNF-α、IL-1β和IL-6基因转录水平的影响。实验结果显示,50、100、200、400、800 μg/cm3 APS可以显著抑制巨噬细胞的氧呼吸爆发(ROS);100、200、400、800 μg/cm3APS可以显著抑制细胞的氮呼吸爆发(NO);50、100、200 μg/cm3 APS预处理后可以显著增强脂多糖(LPS)刺激头肾巨噬细胞的吞噬活性;200 μg/cm3 APS可以显著增强TNF-α、IL-1β和IL-6 mRNA在6、12、24 h的表达量。结果表明,APS对离体培养大黄鱼头肾巨噬细胞有明显的免疫调节作用。

Abstract:
This paper reports the immunoregulation of Astragalus polysaccharide (APS) on macrophages from the head kidney of large yellow croaker (Larimichthys crocea). APS was isolated and purified from Astragalus for the experiment. The effects of respiratory burst and phagocytic activity on the macrophages were detected by flow cytometry. The effects of APS on the gene transcription level of three cytokines TNFα, IL-1β and IL-6 were determined by qPCR. Experimental results showed that 50,100,200,400,800 μg/cm3 APS could significantly inhibit the reactive oxygen species (ROS) of macrophages; 100,200,400,800 μg/cm3 APS could significantly inhibit the reduction of nitric oxide (NO); 50,100,200 μg/cm3 APS could significantly increase the phagocytic activity of macrophages with LPS stimulation in the large yellow croaker. 200 μg/cm3 APS could significantly enhance the expression of TNFα, IL-1β and IL-6 mRNA at 6 h, 12 h and 24 h. The results indicated that APS had a significant immunoregulation on the cultured macrophages from the head kidney of large yellow croaker.
