Abstract:
Taken laminarin extraction rate as an index, the technological conditions of laminarin extraction from laminaria processing wastes were optimized by twostage method of microwave and compound enzyme. When taking laminaria raw material 2.00 g, the optimal technological conditions for the microwave stage were as follow: the microwave extraction solution was pH 6.5, the ratio of liquid to substrate was 60.00 mL/g with 12 min 400 W microwave treating at 60 ℃. Under these optimal conditions, the extraction rate of laminarin could reach 106.49 g/kg. The optimal technological conditions for the compound enzyme stage were as follow: the compound enzyme extraction solution was pH 6.00, the ratio of liquid to substrate was 40.00 mL/g and the enzyme treated for 105 min at 55 ℃ with the dosage of combined enzymes including cellulase 4.00×10
3U, papain 4.00×10
3U and pectinase 2.40×10
3U. Under these optimal conditions, the extraction rate of laminarin could reach 115.16 g/kg. Under the optimal conditions of the microwave and compound enzyme assisted extraction, the extraction rate of laminarin could reach (208.93±0.18) g/kg. The results of antioxidant activity showed that the extracted laminarin had certain antioxidant capacity besides a stronger scavenging activity of hydroxyl (·OH).
